We are seeking a highly motivated bench Scientist to support and develop Affini-T’s proprietary TETHERTM platform for generating highly potent and specific T-cell engagers for solid tumors. The Scientist will have the opportunity to play a critical role in impacting all aspects of the platform including TCR affinity maturation, protein engineering and production, and characterization of T cell engager molecules.
Essential Duties and Responsibilities
The major tasks for this position are as follows:- Design and generate expression constructs for the expression of T cell engagers from mammalian systems
- Express, purify, and characterize novel T cell engagers to support screening and lead optimization in discovery programs
- Construct high diversity libraries and execute TCR affinity maturation campaigns
- Maintain accurate records of inventory, experiments, protocols, and data analysis for documentation and reporting purposes
- Participate in the preparation and delivery of scientific presentations for team meetings
Desired and Required Qualifications
Education and Experience: - PhD in Biochemistry, Biology, or related field, with 1+ year of industry experience, MS with 5+ years of industry experience, or BS with 8+ years of industry experience
- Demonstrated experience with mammalian cell culture required
- Experience with expression construct design and molecular biology (including cloning, qPCR, and next-generation sequencing)
- Experience with assay development to assess immune cell engagers including ELISA, tumor cell killing, cytokine secretion, multi-parameter flow cytometry, and T cell activation assays
- Experience in purification of proteins with affinity, ion exchange, size exclusion, hydrophobic interaction chromatography preferred
- Experience with AKTA FPLC systems for protein purification preferred
- Ability to work well as both team member and as individual contributor
- Excellent communication skills for cross-functional, management and external interactions; excellent oral presentation skills are critical
- Strong organizational skills to ensure thorough and timely documentation of experiments, and preparation of data for compiling reports and presentations to research and corporate teams
